Composition per liter:
Agar ..................................................................14.0g
Sodium citrate...................................................10.0g
Lactose................................................................7.5g
Sucrose................................................................7.5g
Pancreatic digest of casein..................................7.5g
Peptone................................................................7.5g
NaCl....................................................................5.0g
Na2S2O3·5H2O ....................................................5.0g
Yeast extract........................................................3.0g
Meat extract ........................................................3.0g
Sodium deoxycholate..........................................2.5g
Bromcresol Purple ............................................0.02g
pH 7.2 ± 0.2 at 25°C
Preparation of Medium: Add components to
distilled/deionized water and bring volume to 1.0L.
Mix thoroughly. Gently heat and bring to boiling.
Pour into sterile Petri dishes without sterilization. Do
not autoclave. Use the same day.
Use: For the differential isolation of Gram-negative en
teric bacilli. For the isolation of Salmonella, Shigella,
and other nonlactose- and nonsucrose-fermenting mi
croorganisms. Nonlactose/nonsucrose-fermenting mi
croorganisms appear as colorless or blue colonies.
Lactose/sucrose-fermenting microorganisms, such as
coliform bacteria, appear as yellow-opaque white colo
nies surrounded by a zone of precipitated deoxycholate.
