Composition per liter:

MgCl2, anhydrous ...........................................10.93g

NaCl..................................................................7.34g

Casein hydrolysate............................................4.59g

Tryptose ............................................................4.59g

Agar ....................................................................2.7g

KH2PO4.............................................................1.47g

Malachite Green oxalate .................................0.037g

Novobiocin.................................................... 10.0mL

pH 5.2 ± 0.2 at 25°C

Source: This medium is available as a premixed

powder from Oxoid.

Novobiocin Solution:

Composition per 10.0mL:

Novobiocin........................................................0.02g

Preparation of Novobiocin Solution: Add no

vobiocin to 10.0mL of distilled/deionized water. Mix

thoroughly. Filter sterilize.

Preparation of Medium: Add components, ex

cept novobiocin solution, to distilled/deionized water

and bring volume to 990.0mL. Mix thoroughly. Gen

tly heat to boiling. Do not autoclave. Cool to 45°–

50°C. Aseptically add 10.0mL of sterile novobiocin

solution. Mix thoroughly. Pour into sterile Petri dish

es. Air-dry plates for at least 1 hr.

Use: For the isolation and cultivation of motile Sal

monella species from food samples.
