TB Nitrate Reduction Broth

Composition per 100.0mL:

Na2HPO4·12H2O.............................................0.485g

KH2PO4...........................................................0.117g

NaNO3 ............................................................0.085g

pH 7.0 ± 0.2 at 25°C

Preparation of Medium: Add components to

distilled/deionized water and bring volume to

100.0mL. Mix thoroughly. Distribute into tubes or

flasks. Autoclave for 15 min at 15 psi pressure–

121°C.

Use: For the differentiation of Mycobacterium spe

cies based on nitrate reduction. After growth of cells

in appropriate medium, nitrate reduction is deter

mined by making a suspension of cells in TB nitrate

reduction broth and adding hydrochloric acid, sulfa

nilamide, and N-naphylenendiamine. Nitrate reduc

tion turns the medium pink. Mycobacterium

tuberculosis reduces nitrate and turns the medium

deep pink within 1 min. Mycobacterium bovis does

not reduce nitrate and does not change the medium.
