Composition per 4.225L:

Malachite Green................................................0.75g

Egg yolk emulsion ......................................... 3.125L

Glycerol ...................................................... 100.0mL

pH 6.75 ± 0.2 at 25°C

Egg Yolk Emulsion:

Composition:

Chicken egg yolks..................................................66

Whole chicken egg ..................................................6

Preparation of Egg Yolk Emulsion: Soak eggs

with 1:100 dilution of saturated mercuric chloride so

lution for 1 min. Crack eggs and separate yolks from

whites. Mix egg yolks with 6 chicken eggs.

Preparation of Medium: Add components to

distilled/deionized water and bring volume to 1.0L.

Mix thoroughly. Distribute into tubes. Autoclave for

15 min at 15 psi pressure–121°C.

Use: For the isolation of Mycobacterium species.
