Haloferax volcanii Medium

培养基配方

Composition per liter:
NaCl..........................................................................................206.0g
MgSO 4 ·7H 2 O..............................................................................37.0g
KCl................................................................................................3.7g
Yeast tryptone solution ..........................................................100.0mL
Tris[hydroxymethyl]aminomethane·HCl
(1M solution, pH 7.2)........................................................50.0mL
CaCl 2 ·2H 2 O solution..................................................................5.0mL
MnCl 2 ·6H 2 O solution ................................................................1.7mL
pH 7.2 ± 0.2 at 25°C
Yeast Tryptone Solution:
Composition per 100.0mL:
Tryptone........................................................................................5.0g
Yeast extract..................................................................................3.0g
Preparation of Yeast Tryptone Solution:  Add components to distilled/deionized water and bring volume to 100.0mL. Mix thoroughly.

CaCl 2 ·2H 2 O Solution:
Composition per 100.0mL:
CaCl 2 ·2H 2 O.................................................................................10.0g
Preparation of CaCl 2 ·2H 2 O Solution:  Add CaCl 2 ·2H 2 O to distilled/deionized water and bring volume to 100.0mL. Mix thoroughly.
MnCl 2 ·6H 2 O Solution:
Composition per liter:
MnCl 2 ·6H 2 O.............................................................................75.0mg
Preparation of MnCl 2 ·6H 2 O Solution:  Add MnCl 2 ·6H 2 O to distilled/deionized water and bring volume to 1.0L. Mix thoroughly. 

Preparation of Medium: Add components to distilled/deionized water and bring volume to 1.0L. Mix thoroughly. Distribute into tubes or flasks. Autoclave for 15 min at 15 psi pressure–121°C.
Use:  For the cultivation of Haloferax volcanii.
