Composition per liter:

Glucose ....................................................................................... 10.0g

(NH4)2SO4..................................................................................... 6.8g

KH2PO4......................................................................................... 3.0g

Yeast extract.................................................................................. 1.0g

MgSO4 .......................................................................................... 0.5g

CaCl2·2H2O................................................................................. 0.25g

pH 1.65 ± 0.2 at 25°C

Preparation of Medium: Add components to distilled/deionized

water and bring volume to 1.0L. Mix thoroughly. Adjust pH to 1.65

with 50% H2SO4. Distribute into tubes or flasks. Sterilize by heating to

100°C for 30 min.

Use: For the cultivation of Thermococcus acidophilum.
